936 Antigen-presenting cells from HCA2-KO mice are defective in activating regulatory T cells

چکیده

Gut commensal microbes affect the mucosal immune system via expansion of regulatory T cells (Treg) in colon. This is mediated short-chain fatty acids (SCFA), bacterial metabolites generated during fiber fermentation. SCFA are also produced by skin bacteria and may activate resident Treg, activity which diminished certain inflammatory dermatoses. Only upon binding to G-protein coupled receptors, exert their antiinflammatory effects. A relevant receptor for HCA2. Since ligation HCA2 causes suppression intestinal inflammation, we studied whether absence might be responsible recently described impaired Treg HCA2-knock out mice (HCA2-KO). Adoptive transfer experiments isolated from lymph nodes spleens sensitized wild type (WT) HCA2-KO revealed that WT-Treg significantly reduced contact hypersensitivity recipient animals, whereas this effect was not observed recipients HCA2-KO-Treg. To study HCA2-deficiency affects antigen-presenting cells, bone marrow-derived dendritic (BMDC) WT or were examined flow-analysis. BMDC failed release IL-10 aldehyde dehydrogenase 1A involved generation forkhead box P3 (Foxp3) positive Treg. phenotype losing capacity induce prove potency priming coculture performed. These results suggest deficiency alters DC such a way they any longer able induce/activate

Regulatory T Cells in Colorectal Cancer

One of the essential protection mechanisms of immune system is Treg cells which play an important role in maintenance ofimmune homeostasis. However, they may also inhibit immune functions against tumor cells. It has been reported that Treg cellsare increased in patients suffering from different types of cancer. Increased number of Treg cells has been shown in tumor lymphnodes, peripheral blood,...

Low 17β-estradiol Levels Are Better Inducers of Regulatory Conditioned T Cells In-Vitro

Background: 17β-estradiol (E2) has been known to modulate immune response. Recent studies indicate that E2 at pregnancy level plays a role in regulating T cell response. Objective: To investigate the optimum dose of E2 (from 10-9 to 10-7 M) in mediating the generation of regulatory T cells (Tregs), using naïve human CD4+ T cells from healthy women. Methods: Naïve peripheral T cells were purifie...

Circulating Follicular Regulatory T Cells Are Defective in Multiple Sclerosis.

Follicular regulatory T cells (TFR) have been extensively characterized in mice and participate in germinal center responses by regulating the maturation of B cells and production of (auto)antibodies. We report that circulating TFR are phenotypically distinct from tonsil-derived TFR in humans. They have a lower expression of follicular markers, and display a memory phenotype and lack of high ex...

T Helper Cells Profile and CD4+CD25+Foxp3+Regulatory T Cells in Polycystic Ovary Syndrome

Background: Polycystic ovary syndrome (PCOS) is considered as the most common cause of female infertility that affects 4-10% of women in the reproductive age. Previous studies have shown the role of a balanced immune response in a successful pregnancy and fertility. Objective: To investigate the T helper cells type 1 (Th1) /Th2/Th17/Treg paradigms in peripheral blood of infertile PCOS compared ...

Langerhans cells protect from allergic contact dermatitis in mice by tolerizing CD8(+) T cells and activating Foxp3(+) regulatory T cells.

Allergic contact dermatitis is the most frequent occupational disease in industrialized countries. It is caused by CD8(+) T cell-mediated contact hypersensitivity (CHS) reactions triggered at the site of contact by a variety of chemicals, also known as weak haptens, present in fragrances, dyes, metals, preservatives, and drugs. Despite the myriad of potentially allergenic substances that can pe...
